Outline of Annual Research Achievements |
本研究では、多面的機能を備えるmTOR阻害剤の虚血性脳損傷に対する脳保護効果について、マウスくも膜下出血(Subarachnoid haemorrhage: SAH)モデルを用いた検証を実施した。本研究課題実施期間に、論文タイトル"Rapamycin protects against early brain injury independent of cerebral blood flow changes in a mouse model of subarachnoid haemorrhage"がClinical and Experimental Pharmacology and Physiology誌に受理された(Sasaki et al., 2018)。 最終年度はCOVID-19による研究活動の遅延を挽回すべく、高齢化したマウスを用いてEndovascular perforation法によるSAHモデルを作製し、mTOR阻害剤の脳保護効果について検証を行った。現在、その結果について解析段階であるが、mTOR阻害剤を投与する事により、シャム群(血管の穿刺を実施しない群)やコントロール群(mTOR阻害剤を投与しない群)に比較し、SAH後の行動異常や脳虚血からの回復が早い傾向にある結果が得られた。 本検証から、mTOR阻害剤は老化した脳におけるSAHに起因した虚血性脳損傷に対して、脳保護効果を有する可能性があると推察される。本研究成果は治療に難渋する高齢者のくも膜下出血の予後改善に寄与する治療法確立に役立つ基礎的知見となる可能性がある。
