Outline of Final Research Achievements

We have established a novel method for establishing gastric organoid. Gastric organoids were established from 104 gastric cancer cases. By combining a normal organoid and gene editing or Helicobacter pylori or Epstein-Barr virus infection, the model can be used for studying early neoplastic stages. Furthermore, as the organoid model recapitulates the primary tissue status, the GC organoid model can be used to screen drugs to guide patient treatment and to accelerate anti-cancer drug development.

Academic Significance and Societal Importance of the Research Achievements

胃癌は日本人に多い癌であり胃癌制圧の為の基礎研究が急務である。これまでに多くの胃癌ゲノムデータが蓄積されているものの胃上皮培養法の欠如により胃発癌を端的に解析することが困難であり胃発癌分子機構は未解明であった。本研究では新たな胃培養モデルであるオルガノイドモデルの樹立に成功しEBV胃癌モデル構築を試みた。本モデルはEBV胃癌以外の胃疾患研究についても応用が可能であり今後広い分野に渡り胃研究の基盤モデルとなると考えられる。
