Outline of Annual Research Achievements

細胞の遺伝情報はゲノムにコードされており、人体を形成する細胞は全て同一の遺伝情報を持つ。そのため、細胞が固有の機能を獲得し組織形成し個体形成に至るには、選択的な遺伝子発現が不可欠である。この選択的な遺伝子発現を理解するには、(1)転写制御系の解明、転写因子の結合、ヒストン修飾やクロマチン構造変換、RNAポリメラーゼIIの遺伝子座への結合に至る一連のダイナミクスの同時測定、(2)RNA量の測定、(3)翻訳されたタンパク質量の測定に至る多角的な測定を、同一サンプルを用いて行うことが必要である。しかし、各測定はエピゲノミクス、トランスクリプトミクス、プロテオミクス、メタボロミクスのように階層ごとに計測技術が発展してきたため、同一サンプルを用いた共起情報の取得は行われていない。本研究では、各階層を横断的に測定する、すなわちトランスオミクス解析のために、網羅的な抗体を利用したトランスオミクスデータ同時取得法の確立を目指した。2017年度から2018年度の2年間にわたって技術部分の開発を進めた。本研究において主軸となったのは、エピゲノミクス、トランスクリプトミクス、プロテオミクス、メタボロミクスによる細胞内分子数の測定をRNAに変換して検出する技術である。エピゲノミクス部分については、クロマチン挿入標識法の開発に成功し2018年度に報告を行った。トランスクリプトミクス部分についてはCellSeq2法を改良化した独自の手法を樹立し現在投稿準備中である。またプロテオミクス、メタボロミクスについてもクロマチン挿入標識法と同様の手法ができることを確認しており現在検証を進めている。
