Outline of Annual Research Achievements

申請者は、これまでに明らかにしてきた乳がん発現亢進分子BIG3のPKA/PP1αとの三者複合体によるがん抑制因子PHB2の不活化機構に基づき、新たなBIG3複合体による抑制因子の不活化機構を解明して「新たな乳がんの多段階発がん機構の提唱」およびBIG3-PHB2相互作用阻害に加えて、BIG3と新規抑制因子の相互作用阻害剤の開発を遂行して「抑制因子群活性化を利用した休眠抑制因子再活性化創薬」を目指す。当該最終年度では、①各種ストレス依存性PHB2新規リン酸化部位・責任キナーゼの同定を通じたPHB2の意義解明、②BIG3複合体の結合分子探索を通じた新規機能の解明、③BIG3/PHB2相互作用阻害ペプチドについて、PHB2ペプチド開発、④乳がん臨床検体を用いた免疫組織染色と相関解析を本研究期間に遂行した。本年度は、下記の通り、他の項目は概ね遂行できた。
①HER2陽性乳がんにおいて、HER2・EGFR活性化シグナル依存的PHB2リン酸化部位および各責任キナーゼを同定した。さらに、各リン酸化特異的抗体を作製し、免疫細胞染色法・生化学的手法にて、BIG3-PHB2複合体の細胞質およびミトコンドリア局在を明らかにした。
②プロテオーム解析により、数種のミトコンドリアタンパク質をBIG3複合体結合分子として同定し、その新規分子との結合領域の同定およびsiRNAによる結合分子およびBIG3の発現抑制にてミトコンドリア形態異常惹起を明らかにした。
③PHB2はBIG3と2箇所にて相互作用しており、両領域を占めるPHB2由来ペプチドの投与にて、PHB2-BIG3の相互作用阻害を通じた細胞増殖抑制効果を認めた。
④100例のHER2陽性乳がん組織を用いて、BIG3, 各種PHB2リン酸化の発現を検証し、予後と相関関係にあることを明らかにした。
