血小板膜糖蛋白GPIIb/IIIaの細胞外ドメインを分子動力学計算の対象とした。細胞内ドメインのわずかな構造変化が細胞外ドメインの大きな構造を惹起する「分枝レバレッジ仮説」の妥当性検証のために、GPIIb/IIIaの細胞外ドメインの活性型構造と非活性型構造の遷移の可能性を検証した。
分子動力学計算には本予算にて施設内に購入したXeon Pfiおよび学外高性能コンピューターとしてOakforest-PACSを用いた。GPIIb/IIIaの活性型構造に、リガンドとしてフィブリノーゲンのドデカペプチドが結合した構造を初期構造とした。リガンドが結合した条件では2フェムト秒毎に分子動力学計算を行い安定した構造も活性型構造に近似した。「分子レバレッジ」仮説の前提となる細胞外ドメインの複数のエネルギー的安定構造を明らかにするために、ドデカペプチドを外して2フェムト秒毎の分子動力学計算を10,00,000ステップ施行した。計算の継続とともに、細胞外ドメインは、中央付近にて折れ曲り、非活性型構造に近似した形態へと変化した。2年間の研究により、GPIIb/IIIaの細胞外ドメインは、GPIbalphaとは異なり、エネルギー的に安定した構造が複数存在すること、そのエネルギー安定構造の一部は活性型構造に、一部は非活性型構造に類似していること、お互いのエネルギー的安定構造間の遷移を惹起するためのエネルギー障壁は高くないことを確認した。
細胞膜を支点、細胞内ドメインを力点、細胞外ドメインを作動点とした「分子レバレッジ」とするために、細胞膜貫通ドメインおよび細胞内ドメインの構造を、過去に公開されたNMR解析と矛盾しない構造として自作した。「分子レバレッジ」における力点、支点、作用点となるアミノ酸を明確にするための予備的研究にも成功した。
