Outline of Annual Research Achievements

炎症性腸疾患(IBD)の長期罹患例では、大腸の慢性炎症を母地に大腸癌を高率に合併することが知られており、IBD罹患者数の増加に伴い、その予防法及び治療法の開発が望まれている。本研究では炎症性腸疾患合併大腸癌の病態を明らかにすることでその予防法及び治療法を検討する。C57BL6マウスにデキストラン硫酸ナトリウム(DSS)で誘導した大腸炎組織では、腸管へのマクロファージ系細胞の増加とTNFの発現亢進を認め、アゾキシメタン投与後にDSSによる腸炎を3回繰返し誘導した炎症性大腸腫瘍組織では、Tumor necrosis factor receptor 2の発現亢進を認めた。マクロファージ系細胞株RAW264にLipopolysaccharide(LPS)で刺激を行うと、TNFのmRNA及びの膜型TNFタンパクの発現が一過性に上昇、細胞外に遊離型TNFが増加し、4時間後からmRNA及びの細胞表面の膜型TNFタンパクの発現低下を認めた。Tumor necrosis factor-alpha-converting enzyme(TACE)の遺伝子欠損により膜型から遊離型TNFの変換を阻害したところ、非欠損株に比べて膜型TNFの発現亢進を認めたが、やはり発現は一過性で、刺激6時間後から膜型TNFの発現低下を認めた。薬剤スクリニーニングの結果、IkarugamycinがLPSによる膜型TNFの発現亢進・発現持続を誘導すること、遊離型TNFの産生も亢進させることを見出した。TACE欠損RAW264細胞の免疫組織染色では、LPS刺激によりTNFが細胞内に染色される一方でIkarugamycin共刺激では細胞膜でTNFが強発現し、IkarugamycinによるTNFタンパク発現の局在変化を認めた。以上よりIkarugamycinはTNFによる炎症収束メカニズムを阻害することを示した。
