光周波数変調による血流・組織蛍光定量と血液信号抑制

Outline of Annual Research Achievements

ICGを用いた非侵襲的な血流量計測法は申請者が国内特許取得、論文執筆、米国、ヨーロッパ、中国への国際特許申請中である。また、複数の狭帯域レーザー、およびセンサーを応用し、自然光では見えない深部の対象物の観察をしている。特にヘモグロビン信号抑制に特化した「血液信号抑制(blood signal suppression)の実用化を目指す。深部観察に適した吸収の小さな波長を有する狭帯域のレーザー光を照射し、組織からの散乱光を抑制して、血液信号抑制と深部術野可視化を実現する。レーザー光へ熱エネルギーを加えたレーザー(YAG等)など止血操作の基礎実験を行う。1000nm近傍の光源による観察は現在申請中の申請特許が臨床実用化に極めて適している。申請者は近赤外光とICGへの790nmの励起光を組み合わせることで、解剖と血流情報を同時に観察することが可能となり、実臨床応用の論文を投稿中である。脳神経外科顕微鏡から出力される鏡筒と対象物までの距離、拡大率、光源の強さ補正を行ったうえで、蛍光信号を定量化する。また、腫瘍励起光は630nmの狭帯域であり、一方ICG蛍光は可視光外の810nmであるため、単純な自然光との併用は不可能である。このため、複数の狭帯域フィルターの組み合わせ、高速な励起光変換をして融合画像を作成する。
