Outline of Final Research Achievements

Mineralization is predominantly mediated by osteoblasts, which secrete mineral precursors, most likely through matrix vesicles (MVs). Here, we utilize field emission-scanning electron microscopy-based scanning electron-assisted microscopy for assessing live osteoblasts in mineralizing conditions at a nano-level resolution. We found that the calcium-containing vesicles were multivesicular bodies containing MVs. They were transported via lysosome and secreted by exocytosis.

Academic Significance and Societal Importance of the Research Achievements

本研究成果により、骨や歯といった硬組織形成の基本的なメカニズムに関する理解が深まり、骨粗鬆症や歯周病等の硬組織疾患の病態解明や治療法の開発につながることが期待されます。さらに、今回観察に用いた誘電率顕微鏡は培養細胞のみならず、様々な微小物質を溶液中で直接観察することが可能であり、今後も様々な分野での応用が期待されます。
