新型インフルエンザウイルスの出現機構とその制圧

Research Abstract

1.H5N1インフルエンザウイルスの鳥→ヒト及びヒト→ヒト感染に関わる要因の解析
・ヒトから分離した.H5N1ウイルスA/Vietnam/UT3062/04株およびA/Vietnam/UT3028/03株のフェレットにおける致死率は、それぞれ67%および0%であった。これら2株の遺伝子組換えウイルスを用いて、フェレットにおける病原性解析を行ったところ、HA蛋白質とNS遺伝子がコードする2つの蛋白質の合計5カ所のアミノ酸が病原性に関与していることが明らかになった。
・マウスでの病原性が異なるパンデミックインフルエンザウイルスを用いて、病原性に関わる因子を・解析し、PB2とPB1のアミノ酸が病原性に寄与していることを明らかにした。さらに、マイクロアレイ解析を行い、病原性発現に影響を与えている宿主因子を同定した。
2.抗インフルエンザ薬耐性ウイルス
・オセルタミビル耐性のPandemic(H1N1)2009ウイルスについて、ザナミビル、ラニナミビルおよびT-705に対しては感受性を保持していることを明らかにした。
3.粒子形成機構
・電子線トモグラフィー法を用いてインフルエンザウイルス粒子内に取り込まれたゲノムーウイルス蛋白質複合体(RNP)の立体構造解析を行い、8本のRNPが物理的に結合した巨大複合体を形成していることを明らかにした。本成果により、インフルエンザウイルスが持つ分節化ゲノムのパッケージング機構の一端が明らかになった。
