Research Abstract

RhoGTPaseは、アクチンと微小管の動態を制御して細胞形態、運動、接着を調節している。本研究は、RhoGTPaseの下流で働く標的分子の細胞、個体での役割を解析し、Rho情報伝達経路の全体像を明らかにすることを目的としている。本年度は、以下の5点について主な成果を得た。
(1) 細胞質分裂時、mDiaのアイソフォームのうちmDia2は選択的に分裂溝に局在し、機能する。この局在化機構の解析のため、mDia2の結合蛋白質としてAnillinを単離し、mDia2の分裂溝への局在にはRhoとともにAnillinとの結合が必須であることを明らかにした。
(2) mDiaの個体での機能を解析するために、mDia1, mDia3の二重欠損マウスを作出し、その表現型の解析を行った結果、皮質脊髄路の投射異常と嗅球への神経細胞移動の障害を見出した。
(3) mDia1遺伝子欠損マウスより単離したMEF細胞に温度感受性v-Src発現ウイルスを感染させた細胞では野生型細胞と比して、足場依存性増殖、foci形成、in vivoでの腫瘍形成能に著しい抑制を認めた。また、mDia1欠損細胞ではv-Srcの細胞膜への局在化の減弱を認め、このことから、mDia1は、Srcの細胞膜への局在化を制御し、細胞悪性化に寄与していることを見出した。さらに、DMBA/TPA塗布化学発癌実験をmDia1遺伝子欠損マウスに施したところ、本マウスでは野生型と比べて腫瘍形成に顕著な抑制を認めた。
(4) ROCK-I、-II二重欠損マウスは発生初期に致死になること、またホモヘテロ、ヘテロホモの個体はE9.5~E12.5における卵黄嚢上の血管形成が障害され、致死になることを見出した。
(5) マウス排卵時、卵細胞周囲の卵丘細胞が放出するケモカインによりRho/ROCK経路が機能し、インテグリンと細胞外基質との結合を強化し、精子侵入を制御していることを見出した。
