DNAアダクトームによる未知DNA損傷の構造決定とその生物影響評価

Research Abstract

DNA損傷は、発がんや老化の原因であると考えられている。DNA損傷は、外来の紫外線、放射線、発癌物質などによって引き起こされるだけでなく、体内で生じる活性酸素、活性窒素、過酸化脂質などによっても引き起こされる。これまで多くの化学者が様々な発癌物質のDNA付加体の化学構造を決定してきたが、これらのうち、実際の生体中で検出されたものは稀であった。本研究では高速液体クロマトグラフータンデム質量分析器(LC/MS/MS)を用いることにより、様々なDNA損傷を高感度(一億塩基あたり一個のレベル)で正確に定量する方法の開発を進めている。19年度はアセトアルデヒドによるDNA付加体、N2-ethylidine-dGを指標としてヒト血液や実験動物の臓器中の付加体レベルを測定し、アルコール曝露(飲酒)により、この付加体が確実に形成されていること、また、ALDH2の遺伝子多型によってそのリスクが大きく異なることを明確に証明した。また、複数献体からの様々な臓器のDNAにおけるDNA付加体をDNAアダクトームの手法を用いて網羅的に解析した。この結果、DNA付加体の臓器特異性や個人差のデータが蓄積されつつある。また、ヒト臓器中に見られる主要な付加体の構造もいくつか明らかになりつつある。また、既知のDNA付加体の定量法開発も進めており、サフロール付加体、アクリルアミド付加体、胆汁酸付加体などについて検討した。また、新規大気汚染物質3,6-dinitrobenzo[e]pyreneの変異メカニズムや、キノリンのDNA損傷メカニズムについても検討した。
