| Research Abstract |
メラニン色素は皮膚や毛髪を作る細胞自身が合成するのではなく、皮膚の基底層に存在するメラノサイトと呼ばれる特殊な細胞で合成され、メラノソームと呼ばれる特殊なオルガネラに貯蔵されている。核の周辺で成熟したメラノソームは細胞内に張り巡らされた微小管とアクチン線維により、細胞膜まで輸送され、細胞膜につなぎ止められる。メラノソームは最終的に隣接するケラチノサイトや毛母細胞に受け渡され、皮膚や毛髪の暗色化が引き起こされる。このようなメラノソームの細胞内での動きは顕微鏡でも容易に観察することができることから、その動き自身は古くから知られていたが、メラノソーム輸送の分子メカニズムに関する詳細は現在でも十分に解明されていない。本研究課題では、メラノソーム輸送の各輸送ステップ(メラノソームの形成・微小管輸送・アクチン線維への受け渡し・アクチン輸送・細胞膜へのつなぎ止め・ケラチノサイトへの転移の6つのステップ)を制御する因子を同定することにより、メラノソーム輸送の分子メカニズムの全貌を解明することを目的としている。これまでの研究で、アクチン線維への受け渡し及びアクチン輸送に必須の役割を果たす低分子量G蛋白質Rab27Aを不活性化する酵素Rab27A-GAPの同定に成功している。また、本年度は特に微小管上のメラノソーム輸送を司るRabに焦点を当て、マウスやヒトに存在するRabファミリーを網羅的に解析することにより、候補となるRabアイソフォームを同定することにも成功した。
