幼雛期における体温調節メカニズムの解明

Research Abstract

アミノ酸やジペプチドの中に、ニワトリヒナを鎮静化させエネルギー消費を減少させるもの、逆に興奮を促すものを見出した。コルチコトロピン放出因子(CRF)は多彩な生理活性を有し、生命活動を維持するための適応反応に必要なエネルギーホメオスターシスを調節する視床下部-下垂体-副腎皮質(HPA)軸における中枢制御を担っている。中枢にCRFを投与すると酸素消費量、二酸化炭素排出量ならびに熱生産量が高まり、体温の上昇を導く。そこで体温上昇に関わる遺伝子群の発現(avUCR, avANT and avPGC1α)とミトコンドリアにおける生体エネルギー反応に関わる脂肪酸輸送と酸化反応に関する遺伝子群の発現(CPT-I, CPT-II, LCAD, 3HADHおよびCS)とその酵素活性(3HADHおよびCS)に対するCRFの貢献を調査した。まず、熱的中正圏において実験を行ったところ、CRFは幼雛の体温上昇と血漿遊離脂肪酸濃度の上昇を導いた。体温上昇に関わる遺伝子群(avUCPおよびavANT)に対してCRFは有意な効果を有さず、脂肪酸の酸化に関わる遺伝子ならびに酵素活性が組織特異的に高まった。特にCRFは脂肪酸輸送とβ酸化に関係することが判明した。次いで、CRFは寒冷暴露時の体温低下を防ぐことが判明した。この反応においても、avUCPおよびavANTの遺伝子発現に対してCRFは有意な効果を有さず、脂肪酸の酸化に関わる遺伝子ならびに酵素活性が組織特異的に高まった。以上のことから、幼雛時においてはCRFを介する体温調節が脂肪酸の輸送と酸化に依存していることが判明した。
