生殖機能と摂食行動を制御する脳内のエネルギーセンシングメカニズの解明

Research Abstract

本研究の最終的な目標は、糖、脂質、ケトン体の生体レベルを総合的に感知し、摂食と生殖、エネルギー代謝を制御する「エネルギーセンサー」としての上衣細胞の役割を明らかにすることである。特に上衣細胞に内在し、摂食、生殖、エネルギー代謝を制御する脂肪酸やケトン体のセンシングメカニズムについて、明らかにすべく実験を行った。
遊離脂肪酸については、β酸化の阻害剤を第4脳室に投与したところ、性腺刺激ホルモン分泌が顕著に抑制された。また、この反応は視床下部室傍核へのノルアドレナリン合成阻害剤の投与により消失した。これらの結果は、遊離脂肪酸が正のエネルギーシグナルとして後脳上衣細胞に感知され、性腺刺激ホルモンの分泌を制御していること、また後脳上衣細胞からの刺激は視床下部室傍核へ投射するノルアドレナリン作動性神経により仲介されていることが示された。
また、ケトン体のシグナルとしての可能性を検証したところ、ケトン体も同じく後脳上衣細胞に感知され、負のエネルギーとして摂食と生殖をコントロールしていることが示された。ケトン体に関しても後脳上衣細胞で感知された情報は、視床下部室傍核へ投射するノルアドレナリン作動性神経により仲介されていることが明らかとなった。
以上の結果は、後脳上衣細胞が「エネルギーセンサー」として、血中の各エネルギー基質のレベルを感知し、室傍核がセンターとなり、摂食あるいは生殖機能を制御している可能性を強く示唆している。
