ASKファミリーシグナルによる粘膜免疫制御機構の解明

Research Abstract

口腔は,細菌やウイルス感染に対しての最前線の防御システムとして機能している。口腔粘膜の機能の低下ないし破綻は,口腔粘膜疾患だけではなく,感染症をはじめとする様々な全身疾患の発症に直結している。われわれは,ストレス応答シグナルの制御分子であるASK1が,様々な物理化学的ストレスに対する細胞内での情報処理を担うことを明らかにしてきたが,ごく最近になり,ASK1が下流のp38MAPキナーゼを介して,感染初期の迅速な生体防御機構である自然免疫応答において極めて重要な役割を担っていることを明らかにした。昨年度に引き続き,本年度もASKファミリーによる自然免疫制御機構が粘膜免疫においてどのような役割を担っているかを解明することを目的として研究を行い,以下の結果を得た。TLRシグナルによる非病原性細菌認識機構におけるASKファミリーの役割の検討:口腔内や腸内などには多数の非病原性細菌が存在しているが,粘膜免疫機構はそのような常在細菌叢には反応せず,病原体にのみ反応してその排除を行うと考えられてきた。しかし最近,マウスにデキストラン硫酸ナトリウム(DSS)を経口投与することで惹起される腸淡が,TLR2やTLR4のノックアウトマウスでは野生型マウスに比べて重篤になることから,TLRを介したシグナルが常在細菌叢を認識し,粘膜上皮の恒常性の維持や,粘膜損傷に対する保護に働いていることが報告された。
そこで,ASKファミリーノックアウトマウスについてもこのDSS誘導性腸炎に対する感受性を個体,組織両レベルで検討し,新たな粘膜免疫系の機能としても捉えられるこの機構へのASKファミリーの深い関与を明らかにした。
