Research Abstract

ヒト常染色体優性夜間前頭葉てんかん(ADNFLE)責任遺伝子(nACh receptor遺伝子変異 : S284L)を導入したラット(S284L-Tg)は、脳波解析、行動解析、電気生理的機能解析の結果、Face validity(疾患とモデルの表現型の妥当性)、Construct validity(疾患とモデルのメカニズムの妥当性)、Predictive validity(疾患治療薬がモデルにも有効)の3点をクリアする世界初のてんかんモデルラットであることを示し、本年度論文として報告した(J. Neurosci 28 (2008) 12465-12476))。さらにこのモデル動物を用い、細胞レベルにおけるGABA作動性神経の機能異常が、phenotypeとしてのてんかん発作発症以前に生じていることを明らかとした。さらにこのGABA作動性神経異常を発作発症前より是正することにより、治療効果が見られた。このことからは、てんかん根治療法の可能性を示した。また、他の責任遺伝子を導入したラットの詳細な行動解析により、けいれん発作のみならず、ストレス耐性の低下が見られた。この結果からは、本研究による動物作成はてんかんをはじめとした発作関連疾患の病態解明、治療方法開発への応用のみならず、他の精神神経疾患の病態解析への応用も可能とした。上記のヒトの遺伝子変異を導入した遺伝子改変動物モデル以外にも、生体内の機能分子の可視化を目的とした遺伝子改変動物の作成も試みており、その目的遺伝子のプロモーター部分下流に可視化のための蛍光蛋白が挿入された動物作出にも成功した。この可視化遺伝子改変動物の系統確立を現在行っており、病態モデルとしての利用を可能とした。
