Research Abstract

個体発生時の肝臓形成を模倣したマウスES細胞から肝組織へ分化誘導する我々独自の手法を応用して、肝細胞チップではなく、肝組織チップを開発することが本研究の目的であった。胚様体またはES細胞由来肝組織を培養できる蓋開閉型マイクロ培養装置の開発を行い、肝機能を測定することに成功した。また、ES細胞由来肝組織のほか、内皮細胞ネットワーク管状構造の周囲に初代培養肝実質細胞が接着する類洞肝組織構造のマイクロ培養装置の開発にも成功した。以上のことから、肝組織チップが薬物代謝や毒性試験へ応用できる可能性を示すことができた。
