微生物rRNAの自然レベル放射性炭素分析に基づく海洋堆積物中炭素サイクルの解明

Research Abstract

本研究の目的は、堆積物試料からの微生物rRNA等の有機分子レベル放射性炭素同位体比分析システムの開発である。本システムでは、大量のサンプルを必要最小限の前処理だけで、堆積物中の微生物rRNA等の有機分子について、現場に生息する微生物の炭素源に関する一次情報を短時間短時間で得ることを目的とする。平成21年度は、rRNAを変わる微生物起源分子である古細菌細胞膜脂質分子(GDGTs分子に着目し、海水試料からの大量抽出。濃縮・精製のための試料採取法、分析化学的検討などを行い、最終的にGDGTs分子の放射性炭素分析に成功した。古細菌(大きさ平均0.5-0.2μm)細胞膜由来テトラエーテル脂質(GTGTs)の放射性炭素を測定するために必要な海水量は10万-20万Lと試算される。深層海水中の古細菌を補足するため、0.2-0.5μmサイズフラクションを効率的に大量ろ過できる装置を製作した。静岡県駿河湾深層水取水供給施設(397m)において、本システムの大量ろ過試験並びに実試料のサンプリングを実施した。GDGTsは、0.2μmメンブレンフィルターで回収した。その後、タンパク質を特異的に分解する酵素を用いて古細菌細胞膜を分解し、溶媒抽出(ソックスレー抽出等)によりGDGTsを含む溶媒可溶成分を得た後、シリカゲルカラムクロマトグラフィでGDGTsを含むステロール・アルコールフラクションを分離、GDGTsをLC/MSで同定・定量を行った。GDGTs抽出に関する検討では、rRNA同位体分析への条件検討の基礎となる知見を大いに得ることができた。
