量子化学計算による生物無機化学の新たな展開

Research Abstract

密度汎関数計算およびQM/MM計算により、ビオチンカーボキシラーゼ、チロシナーゼ、膜結合型メタンモノオキシゲナーゼの構造と反応機構の研究を行った。特筆すべき実績として、チロシナーゼによるチロシンードーパキノン転化反応に関するものがある。杉山らにより得られたチロシナーゼのX線結晶構造をもとに大規模な計算モデルを構築し、エネルギー論的に有利な反応経路の探索研究を行った。チロシナーゼのX線結晶構造をもとに計算モデルを構築した。まず、酵素反応とは直接関係しないORFタンパクと硝酸イオンは除外した。次に、X線結晶構造には水素原子が含まれていないため、CHARMMプログラムを用いて水素の添加および構造緩和を行った。以上の方法で作成した酵素の原子数は約4300個であり、活性サイトの原子数は約100個である。酵素全体を、量子力学計算のみで計算するのは非常に困難である。そのため、量子力学計算(QM法)と分子力学計算(MM法)を組み合わせたQM/MM法の一種であるONIOM法を用いた。MM領域の分子力場としてAmber力場を用い、QM領域はB3LYP法を用いた。はじめに、チロシンからプロトン移動が起こって活性サイトに結合し、その後、O-O結合の開裂が起こるこの過程が反応の律速段階になると予想される。さらに生じたフェノキシルラジカルと架橋酸索が結合しC-O結合ができる。最後にプロトン移動がヒスチジン配位子を介して起こり、ドーパキノンが生成する。
