管状マスチゴネマの起源の探索とその多様性から見たクロミスタ系統群の進化系統

Research Abstract

黄金色藻(ストラメノパイル系統群)オクロモナスOchromonas danicaより単離・同定された管状マスチゴネマ関連遺伝子Ocm1およびそのファミリーに属するOcm2,Ocm3のオーソログ(cDNAおよびゲノム配列)をストラメノパイル系統群に属する5綱9目11種(褐藻綱:Ectocarpus siliculosus,Hincksia conifera,Desmarestia aculeatea,Saccharina japonica,Sporochnas bolleanus,Halosiphon sp.シゾクラディア藻:Schizocladia ischiensisペラゴ藻:Ankyrochrysis sp.ラフィド藻Heterosigma akasiwoビコソエカ類:Cafeteria sp.)において決定した。決定された配列の解析の結果,これらの遺伝子は非常に速い進化速度をもつが,とくに褐藻綱などにおいてその配列およびイントロン構造が系統解析における有用な分子マーカーに成りうることが示された。また,クリプト藻類Pyrenomonas helgorandii(SAG)の大量培養を行い,pHショック法を応用した鞭毛の単離法を確立した。また,鞭毛の膜タンパク質分画の二次元電気泳動解析により,マスチゴネマのものと思われる幾つかのスポットを特定した。
