Research Abstract

主要膜リン脂質の一種であるスフィンゴミエリン(SM)の生合成では、小胞体で合成されたセラミドが、ゴルジ体に移行してSMへと変換される。我々は、SM生合成に異常を有する哺乳類培養細胞変異株を複数分離し、その中にセラミド輸送欠損変異細胞を見出し、さらに、当該変異株の相補遺伝子をクローニングするという手段によって、セラミドの小胞体-ゴルジ体間選別輸送に特異的に関わる分子装置(CERTと命名)の同定に成功した。
我々はCERT自身がリン酸化修飾されていることを見出し、リン酸化部位の決定とリン酸化が担う役割を明らかにすることを目的とした。
