Research Abstract

植物は土壌中から必須栄養素である鉄を根の細胞膜に存在するトランスポーターによって細胞内に取り込み、必要とされる部位に送り込む。植物は鉄欠乏条件下で鉄の吸収・利用機構に関与する遺伝子群の発現を誘導するが、この遺伝子制御の分子メカニズムはほとんど解明されていなかった。そこで我々は、これまでに同定した鉄欠乏応答性シスエレメントIDEに特異的に結合する転写因子の探索を行った結果、イネの新規転写因子IDEF1 (IDE-binding factor 1)の同定に成功した。IDEF1は植物固有の転写因子ファミリーABI3/VP1の未だ解析されていないサブグループに属し、IDE1内のCATGC配列に特異的に結合するという新規の配列認識特性を持つことが明らかになった。35Sプロモーターの制御下でIDEF1を恒常的に高発現させた形質転換タバコでは、鉄欠乏根のみでIDE1を介した発現が活性化された。鉄欠乏誘導性プロモーターの制御下でIDEF1を発現させた形質転換イネは、鉄欠乏水耕液で鉄欠乏クロロシスの進行が遅く、石灰質アルカリ土壌で発芽させると良好な初期生育を示した。この形質転換イネは鉄欠乏条件下で二価鉄トランスポーター遺伝子OsIRT1と、鉄欠乏誘導性転写因子遺伝子OsIRO2の発現を促進させた。これらの結果から、IDEF1は鉄欠乏応答に関与する多段階の遺伝子制御ネットワークの初期段階を担うことによって鉄欠乏応答性と耐性を制御することが明らかになった。
