Research Abstract

高血圧、高脂血症等,糖尿病に伴う動脈硬化、血管病変、血管傷害に伴うリモデリングのメカニズムなどの解明は、種々の臓器障害の予防、治療のうえで重要な課題であり、レニンーアンジオテンシン系がこれらの病態に、密接に関与していることはよく知られている。アンジオテンシン(Ang)IIは強力な昇圧物質であるばかりではなく、循環器系、中枢神経系、生殖器系等、生体内で幅広く作用し、細胞の増殖、肥大、分化等の重要な機能に深く関わっていることがしられている。AngIIの作用は、主として、AngIIタイプ1受容体(AT1受容体)を介するものと理解されてきたが、申請者の堀内等がクローンしたAngIIタイプ2受容体(AT2受容体)が、血管障害、心筋梗塞後の心血管リモデリング等に、特異的に発現することが知られてくるに従い、AT1受容体とAT2受容体の発現バランスが、これら病態にとって、重要であると考えられるようになってきた。AngII受容体タイプ1(AT1)、タイプ2(AT2)受容体は、互いに拮抗して、心血管リモデリングを調節していることが明らかとなってきた。我々は、AT2受容体のC-末端に特異的に結合するATIP(AT2Receptor Interacting Protein)をクローニングし、ATIPの血管リモデリングにおける作用をATIP過剰発現マウスを作製し検討した。血管傷害において、野生型マウスに比べ、血管平滑筋増殖、炎症、酸化ストレス、新生内膜の減少等が認められ、ATIPは血管保護的に作用することが明らかとなった(Hypertension,2009,in press)。本研究は高血圧、糖尿病に伴う血管病変に対する新しい分子標的の解明に貢献するものであり、今後、ATIPの調節機構を解明することで、高血圧、さらには当尿病の臨床医学の発展にも寄与する意義深い研究である。
