組織特異的ノックアウトマウスを用いた乾癬発症機序におけるSOCSファミリーの研究

Research Abstract

表皮特異的SOCS3欠損マウスを用いた研究:表皮特異的に活性化STAT3を持続的に発現させたマウスにおいて乾癬様病変が出現することが報告されている。SOCS3はSTAT3のnegative regulatorであり、STAT3により誘導される。従って、SOCS3が表皮特異的に減少あるいは消失することにより、STAT3の持続的活性化が誘導され、乾癬様病変が誘導されることが予想される。以上の考えに基づき、表皮特異的SOCS3欠損マウスを用いた研究を行った。K5-CreマウスとSOCS3-flox/floxマウスを掛け合わせることにより表皮特異的SOCS3ヘテロノックアウトマウスを作製し、RT-PCRにてSOCS3がヘテロ欠損していることを確認した。このSOCS3ヘテロノックアウトマウスとSOCS3-flox/floxマウスを掛け合わせることにより表皮特異的SOCS3ノックアウトマウスを作製した。表皮特異的SOCS3欠損マウスの大多数において、後頭部から背部にかけて乾癬様の病変が30週以降に形成された。引き続いてテープストリッピングによる皮膚の刺激試験を行った。脱毛クリームにて背部の毛を脱毛後2日目に3Mテープで20回背部の皮膚を、テープストリッピングを行い、皮膚表面の観察に加えて経時的にサンプルを回収した。SOCS3欠損マウスは皮膚の肥厚、炎症の持続がみられ、乾癬様病変を形成した。組織学的検索では、表皮はコントロールと比較して著明に肥厚しており、STAT3のリン酸化が多数の細胞で認められた。また、増殖のマーカーであるKi67陽性細胞はコントロールと比較して著明に増加していた。以上の結果から、SOCS3ノックアウトマウスは乾癬のモデルマウスとしての可能性を有していることが示唆された。
