| Research Abstract |
スキルス胃癌の病態機序に立脚して、新しい分子標的治療法の開発を目的とした。スキルス胃癌は線維芽細胞の増生を伴った増殖進展を特徴とすることから, スキルス胃癌細胞の増殖進展には間質線維芽細胞の産生する増殖因子FGF-7が癌細胞のFGF-R2/K-samIIに作用していることを明らかにしたし、さらにスキルス胃癌細胞の転移にはTGF-βが関与することを明らかにした。これらの研究により、間質細胞の産生するFGF-7やTGFβがスキルス胃癌細胞の増殖進展に関与し、そのリガンドであるFGF-R2やTGFβRがスキルス胃癌細胞に強く発現していることが重要であることを明らかにした。これらのことからFGF-R2(K-samII)阻害剤Ki23057やTGFβシグナル阻害剤A-77のスキルス胃癌の増殖進展に対する有用性および、抗癌剤との併用効果を明らかにし、新規分子標的治療剤を本邦で開発することを計画した。現在いくつかの分子標的治療薬が悪性疾患に臨床応用されてきているが、胃癌に関しては分子標的治療薬の臨床応用は国内外において認められない。従って、今回検討する分子標的薬剤は胃癌の治療薬として新しい開拓面を有する研究と位置付けられる。
