Research Abstract

1.オリゴDNA/RNAに任意の配列,組成を持つペプチドをコンジュゲートすることに成功した。
2.オリゴDNAにSV40ラージT抗原由来の核局在化シグナルペプチドをコンジュゲートさせ,ヒト白血病細胞由来の株化細胞に対して細胞内への取り込みを共焦点走査型顕微鏡により観察したところ細胞核へ局在化していることが認められた。
3.オリゴDNA-核局在化シグナルコンジュゲートにより,ヒト白血病細胞に発現するテロメラーゼを強く阻害することに成功した。
4.オリゴDNAおよびsiRNAにHIV-1 revタンパク由来核外輸送シグナルペプチドをコンジュゲートさせ,ヒト白血病細胞由来の株化細胞に対して細胞内への取り込みを共焦点走査型顕微鏡により観察したところ細胞質内へ局在化していることが認められた。
