Research Abstract

1. Identification of ncRNA genes responsive to environmental stress.
We searched for low-CO_2-inducible ncRNA genes in the cyanobacterium Synechococcus elongatus PCC 7942, using DNA microarrays that have oligonucleotide spots for both strands of ORFs and intergenic regions. Ten intergenic regions, whose transcription levels were high and whose expression patterns were different from'those of the adjacent ORFs, were selected as candidates of the regions harboring the ncRNA genes. In two of these regions, low-CO_2-inducible small transcripts were detected by Northern blot analysis, confirming the occurrence of ncRNAs induced by CO_2 limitation.
2. Mechanisms of induction of the ncRNA genes.
The transcription start points of the two ncRNA genes were determined. The deduced promoter region of one of the ncRNA genes had an HLR motif typical of high-light responsive genes as well as a CbbR motif (TnA-n7-TnA) found in many low CO_2-resonsive genes. This ncRNA gene was shown to respond to high light as well as low CO_2. The other ncRNA gene had no typical promoter elements nor known cis-acting elements for low-CO_2-induced genes.
3. Role of the ncRNAs in adaptation of cyanobacterial cells to CO2 limitation.
Using an expression vector, the complementary strands of the ncRNA were overexpressed in Synechococcus elongatus cells. The transformants grew slower than the wild-type strain or the strain harboring the empty vector under low CO_2-conditins. The results suggested that the ncRNAs help growth of the cyanobacterium under CO_2- limited growth conditions
