Research Abstract |
We previously reported that augmentation of interleukin (IL)-12 production or intranasal IL-12 administration alleviated influenza pneumonia in mice. In this study we characterized IL-12 production as an early host immune defense against influenza infection. In the bronchoalveolar lavage fluids prepared from intranasally influenza virus-infected mice, the IL-12 level was significantly elevated on day 1, and the production of tumor necrosis factor-a, IL-18, interferon-a, -B, and-Y increased on day 2. Immunohistochemical analyses of the infected lungs on day 1 detected IL-12 and its transcripts in subepithelial smooth muscle beneath the infected bronchiolar epithelium. Macrophages were observed on the bronchiolar epithelium and localized beneath the bronchiolar smooth muscle coat and in parts of the bronchioles where influenza virus antigen was distributed. Biochemical and immunohistochemical analyses revealed IL-12 production in macrophage-like cells near the infected epithelium on day1 and production of the other cytokines in the infected lung thereafter. Thus, IL-12 was produced by macrophages infiltrating near the infected epithelium as the first response cytokine, and IL-12 production around infected area may direct an early immune defense to alleviate influenza infection.
