Research Abstract |
Human intestinal-like cell line C2BBe1 cells expressed higher levels of BCMO1, CRBPII and other vitamin A-related genes' transcripts than parental Caco-2 cells. Human CRBPII gene transcription was synergistically stimulated by HNF-4α and intestinal-specific transcription factors (CDX2 and GATAs). Human BCMO1 gene transcription was competitively regulated by HNF-4α and HNF-1α on the same distal promoter element. Moreover, PMA significantly decreased CRBPII and BCMO1 gene expression levels through PKC-ERK-mediated down-regulation of nuclear HNF-4α and HNF-1 levels in C2BBe1 cells. These transcription factors are also involved in the differentiation-induced vitamin A and β-carotene absorption and metabolism in the human small intestinal cells. In conclusion, HNF-4α and/or HNF-1α may be pivotal transcription factors involving in the bioavailability and homeostasis of human small intestinal vitamin A and β-carotene.
