腎ネフロン特異的HGF受容体ノックアウトマウスを用いた腎再生分子機構の解析

Research Abstract

本年度はHGFが腎組織修復後の再生、修復を担う生理的因子であるとの仮説の元に、生体内HGF受容体/Metの機能を阻害する目的で以下の解析を進めた。
1)急性腎不全マウスでのHGF中和抗体によるMetシグナル阻害
腎虚血により急性腎不全を惹起したマウスにおける腎修復過程での内因性HGFの産生動態を解析した。その結果、腎および肺や肝臓、脾臓といった臓器でHGF mRNAレベルは腎虚血後3-6倍の増加を示した。これに一致して血中HGFレベルは3-5倍に増加した。そこで、HGF中和抗体を腎虚血マウスに投与したところ、Metシグナル伝達阻害とともに腎尿細管上皮細胞の再生は阻害され、腎機能不全の時間が明らかに延長した。HGF中和群では腎組織虚血の持続に一致して、虚血時に誘導されるモノカインSDF-1の発現亢進に一致し、上皮再生阻害を補うように骨髄幹細胞の動員が亢進した(投稿論文作成中)。
2)腎特異的Met/HGF受容体ノックアウトマウスの作成
Met-ATP結合領域(エクソン16)にfloxカセットを挿入したノックインマウスを作製した。次に腎糸球体ポドサイトに特異的にCreを発現するマウスを導入し、両系統を掛け合わせたダブルミューテーションマウスを作製した。このマウスでは腎糸球体特異的にMet ATP結合領域が欠失していることが確認された。
3)エンドトキシン誘発急性腎不全マウスでのHGFによる救命効果
エンドトキシンショックを惹起したマウスは腎ネフロンの傷害により80%以上が4日以内に死亡する。このモデルにリコンビナントHGFを投与すると、ほとんどの動物が生存し、腎ネフロン傷害も軽度であった。
