Research Abstract

イオンスプレー質量分析法(ESI-MS)は分子構造を知るには最も適した方法であるが、分子が強く荷電していたり、反対に全く荷電していないと検出できないという欠点がある。例えば有害な種々の重金属イオンは強く+に荷電し、青酸イオン、アザイドイオン等は強く-に荷電しているので検出できない。生体内では多くの薬毒物は錯体となっているので、錯体の分子構造解析や定量は重要であるが、上記理由により、ESI-MSによる錯体の高感度解析は殆どなされていなかった。申請者は、薬毒物を錯体化し、さらに第三の因子を反応させてternary complexを作成(励起)し、分子を弱く荷電させることによりイオン化を促進し、ESI-MS で高感度に検出する方法を見出した。この励起化法を用いた薬毒物の高感度定量と作用機序の解明が目的である。
(1) モリブデン、プラチナ、クロム、コバルト等の陽イオン含有薬毒物測定のための励起化法の開発
(2) 青酸カリ、アジ化ナトリウム、チオシアン等の陰イオン含有薬毒物のための励起化
(3) ヒ素、セレン等の非金属元素含有薬毒物のための励起化
(4) 解毒に関与するモリブデン酵素と薬毒物中毒との関係解明
(5) 以上を総合した、ESI-MSに適した錯体励起化メカニズムの解明と多くの物質に適した励起化法の開発
