炎症性腸疾患における腸管上皮細胞間接着蛋白の研究

Research Abstract

潰瘍性大腸炎、クローン病、対照大腸組織を用いて、細胞間接着蛋白の免疫染色法における局在を検討した。細胞間接着蛋白に対する抗Rab13抗体、抗VASP抗体、抗リン酸化VASP抗体を用いて検討した。Rab13は、正常では被蓋上皮の最も管腔側にライン状に染色された。クローン病患者では非炎症粘膜においても被蓋上皮の最管腔側にはライン状には染色されず、細胞質側へdislocationしていることが判明した。また、被蓋上皮よりも腺窩上皮が優位に染色される。VASPはRab13によりコントロールされるTJ蛋白であるが、VASPにおいても同様の結果であった。リン酸化VASPについては現在優位な差はでず、引き続き症例数を増やし、検討中である。さらにウサギ抗-tumor necrosis factor(TNF)alfa抗体、ウサギ抗-transforming growth factor(TGF)beta抗体、およびウサギ抗-interferon(IL)6、10、12抗体を用いてfluorescein-conjugatedヤギ抗ウサギIgGF(ab)2affinity purified抗体(ICN Pharmaceutical、,Inc.Ohio USA)を用いた免疫染色をおこない細胞間接着蛋白の局在とサイトカインとの関連を検討している。
