炎症性腸疾患における腸管上皮細胞間接着蛋白の研究

Research Abstract

クローン病、潰瘍性大腸炎、対照大腸組織を用いて細胞間接着蛋白の免疫染色法における局在を検討した。細胞間接着蛋白に対する抗Rab13抗体、抗VASP抗体、抗リン酸化VASP抗体、抗ZO-1抗体を使用した。正常粘膜ではVASP、ZO-1は被蓋上皮の最も管腔側にライン状に染色された。Rab13は被蓋上皮の最も管腔側及び核上部にライン状に染色された。クローン病では、ZO-1、VASP、Rab13ともに細胞質側へのdislocationをみとめ、Rab13、ZO-1は杯細胞内の粘液も染色された。潰瘍性大腸炎ではdislocationは認められず、正常粘膜と同様に染色された。リン酸化VASPは正常粘膜、炎症性腸疾患ともに染色されなかったが、これは組織の固定による影響と考えられる。これらより、クローン病において、細胞間接着蛋白であるRab13、VASPのdislocationはクローン病の腸管粘膜の透過性亢進に関与し、クローン病の病態にも関与していることが示唆された。
