Research Abstract |
1. Sample collection : 714 schizophrenia and 200 bipolar disorder patients samples were collected. Combined with corroboratory groups samples, total sample numbers account more than 1 000 for schizophrenia and 800 for bipolar disorder, respectively. For pharmacogenomics analysis, 120 schizophrenics who were evaluated the efficacy and adverse effect for treatment of atypical antipsychotics risperidone was collected. Also 100 major depressive disorder patients who were evaluated the efficacy for serotonin reuptake inhibitor Fluvoxamine and personality traits were collected. 2. Expression analysis : To determine the stage for analysis, preliminary expression analysis showed immediate moment to establish cell line is most suitable for analysis. 3.30 vs. 30 case and control expression analysis showed three specific genes could distinguish between case and controls for about 83% accuracy by using BFCS method. Consequently we chose top 10 genes using the other selection method S2N' which focus on the differences of discrimination degree. Simulation analysis select 4 genes and the accuracy of this method showed about 78%. We conclude that using expression profile in peripheral blood cell cold be useful to establish diagnostic method. To apply for clinical use, further investigations are needed in larger samples.
