Research Abstract

Angiogenesis is required for tumor growth and progression. We have recently found that pigment epithelium-derived factor (PEDF) inhibits growth of both endothelial cells and tumor cells in vitro, thus suggesting that PEDF could exert anti-tumor effects. In this project, we investigated which PEDF domains could have anti-angiogenic properties both in vitro and in vivo. In vitro, three PEDF peptides (amino acids 381-387, 388-393, 394-400) were found to have anti-angiogenic effects on endothelial cells; their anti-angiogenic potencial was similar to that of PEDF full protein. Further, the peptides had growth inhibitory effects on osteosarcoma cells. In vivo, the peptides were also found to suppression angiogenesis elicited by alkaline injury in cornea. The present study demonstrated that the PEDF peptides had anti-angiogenic and tumorstatic effects on osteosarcoma cells. The observations suggest the potential therapeutic utility of the PEDF peptides for the treatment of osteosarcoma.
