Research Abstract

破骨細胞前駆細胞の蛋白を抽出したところCTR mRNA 3'非翻訳領域(CTR 3'UTR)配列RNA プローブと結合性を持つ蛋白が確認された。結合蛋白の分子量は40-45kDa でAUF-1、HuR関与の可能性を検討した。HA-tag付加AUF1 をNIH3T3細胞に導入し、抽出蛋白と 3'UTR RNA を相互作用させたところ結合性が確認できた。抗HA抗体を利用した免疫沈降で蛋白RNA複合体を集め、結合RNAをRT-PCR法で解析したところ、CTR 3'UTRに設定したプライマーで特異的に増幅された。RNA認識部位を変異させると3'UTR配列との結合性は見られず、蛋白RNA結合特性に依存していると思われ、この現象が受容体制御と関連している可能性が考えられた。
