Research Abstract

現在のヒト卵巣組織の凍結保存技術には様々な克服すべき問題点があるが、悪性腫瘍の治療により生殖機能の傷害が懸念される女性にとって切実かつ喫緊の課題である。また、直接の研究対象となる性同一性障害患者にとっても、治療の選択肢と可能性を拡大する上で、決して小さくない意義を有すると考えられる。
性同一性障害患者より切除された卵巣は通常の組織学的検査に用いられる以外は廃棄されていたが、本研究では余剰部分から豊富な卵巣組織の提供を受け、種々の方法や実験条件で卵巣組織の凍結・融解を行い、残存卵子数の算出およびアポトーシス関連遺伝子群の発現を検討することで、ヒト卵巣にとって最適な凍結・融解方法の確立を目指す。
また、ヒト卵巣組織を免疫不全SCIDマウスに異種移植し、一定期間後の卵巣組織を同様に検討することで、ヒト卵巣にとって最適な条件の確立を目指す。
更に、ヒト卵巣組織の体外培養を行い、これまでに知られている種々の卵子生存因子を培養系に添加し、一定期間の培養後の卵巣組織を同様に検討することで、ヒト卵巣にとって最適な体外培養系の確立を目指す。
