Research Abstract

本年度は実施計画に記載した以下の4つの研究を実施した。(1)ヒトコンデンシン立体構造解析と相互作用解析、(2)HP1BP74のクロマチン高次構造における役割解明、(3)HPlBP74のin vivoにおける相互作用解析のためのベクター構築、(4)ヒストンシャペロンNAP-1の会合状態解析である。(1)はコンデンシンヒンジ2量体の2量体形成を質量分析装置により確認後・結晶化を行い,良好な結晶化を得ることに成功した。得られた結晶を用いてSPring8において測定を行ったところ、分解能2.5A程度の回折像を得ることに成功した。これまでに報告されている結晶構造を用いての位相決定は不可能であったため、現在、SeMe置換体の作製を行っている。一方、Mycタグを付加したヒンジを細胞内で発現させ、免疫沈降によりコンデンシシ複合体を単離することにより、各サブユニット間の相互作用状態を明らかにすることに成功した。(2)についてはヒト組換ピストンを大量調製し、ヌクレオソーム再構築を行い、そこに組換HPIBP74を添加することにより検討を行った。ピストンHlと同様にクロマトソームが形成されればクロマトソームストップが観察されることとなる。その結果、HPIBP74がピストンHlと同様の位置に結合し、クロマチン高次構造形成に寄与していることが分かった。次に、TAP(Tandem Affinity Purification)システムを用いた(3)HPlBP74の免疫沈降を行うために、TAPベクターの入手、およびHPlBP74の挿入を行った。すでにベクターの構築は完了したため、来年度はTAPシステムによる免疫沈降、質量分析による同定を行い、HPIBP74を中心とした相互作用ネットワークを明らかにする。(4)については、米国の研究者との共同研究により超遠心分析を実施し、これまでに報告されていない会合状態を明かとすることが出来た。
