受容体型チロシンキナーゼを標的としたEGCG大腸発癌抑制の研究

Research Abstract

本研究は,緑茶カテキンEGCGの癌細胞増殖抑制効果について分子生物学的な考察に基づいて検討し,最終的には臨床の場において,EGCGやその他のポリフェノールを用いた大腸癌予防の実践を目指すことを目的としている。我々は今年度の研究で,EGCGが腫瘍細胞から産生される増殖因子IGF-1/2の発現を抑制し,IGF-1/2のレセプターで受容体型チロシンキナーゼの一つであるIGF1-Rとその下流分子(ERK,Akt,Stat3)の活性を阻害することで,肝癌細胞にアポトーシスを誘導し増殖抑制効果を発揮することを明らかにした。IGF/IGF1-Rシステムの異常活性は,各種の肥満関連発癌に深く関与しているが,我々はEGCGが同システムの活性を阻害するとともに,高血糖・高インスリン血症・高脂血症・高レプチン血症を改善することで,肥満マウス(db/dbmice)の大腸化学発癌を抑制することも明らかにした。またEGCGが腸管の炎症を改善し,C0X-2やTNF-a,IFN-g,IL-6,IL-12,IL-18といった炎症性サイトカインの発現を抑制することで,DSS/AOM誘発による腸炎関連大腸化学発癌を抑制することも報告したが,これらin vivoにおけるEGCGの大腸発癌予防効果は,すべて生理的な投与量(0.01%EGCGの飲水投与)で確認された。受容体型チロシンキナーゼの異常活性,肥満,慢性炎症は,大腸や肝発癌における重要なrisk factorであることより,EGCGがこれら肥満および腸炎関連大腸発癌を生理的濃度で抑制したことは,今後EGCGやその他の(緑)茶抽出物を用いた大腸や肝発癌予防の可能性を考える上で,重要かつ有意義な結果と考えられた。
