Outline of Annual Research Achievements |
In the last eight months Dr. Sekulic combined behavioural, immunohistochemical and genetic approaches to induce new memories in mice and subsequently label active cells, specifically interneurons involved in the theta rhythm, representative of the novel neuronal ensemble. He evaluated the overlap of the newly marked neurons with the population of inhibitory interneurons of interest, specifically O-LM cells. The mice used were a mix of wild-type, Somatostatin-Cre (SST-Cre) mice for labeling O-LM cells and cfos-tTA and cfos-tTA/SST-Cre double positive mice for marking active neurons during new memories that might be O-LM cells. Additionally, the RAM (Robust Activity Marking) virus, a recently developed tool for marking newly active neurons, was also used in combination with SST-Cre mice. These efforts are an entirely new line of inquiry in my laboratory and I am very satisfied with his efforts and productivity. In parallel Dr. Sekulic has started his training in in vivo physiological recording to monitor the dynamic activity of this neuronal population in freely mice. He has made excellent progress and will soon begin to collect the necessary data.
