Outline of Annual Research Achievements

抗がん剤によるがん細胞のプログラム細胞死(Programmed cell death (PCD))の誘導と制御は、抗がん活性向上や副作用低減などに重要である。これまで申請者は、水中で安定なシクロメタレート型イリジウム(Ir(III))錯体―カチオン性(塩基性)ペプチドハイブリッド(IPH)を設計・合成し、違うペプチドをもつIPHが異なるタイプのPCDを誘起することを報告した。そこで本研究では、光学活性な多脚アロイルチオウレアを導入したルテニウム(II) (Ru(II))-アレン錯体の設計・合成と抗がん活性測定を行った。様々な配位子を導入した光学活性Ru-アレン錯体の合成と構造決定を行った。また、カチオン性(塩基性)ぺプチドを導入したIPHについては、がん細胞(ヒトT細胞性白血病Jurkat細胞)のparaptosisを誘導し、死細胞中で強く発光することを見出した。さらに、IPHが(i)IPHのparaptosis誘導活性には塩基性ペプチドが重要であること、(ii)ペプチド配列やIr錯体中心とペプチド間のリンカー長に依存すること、(iii)IPHがCa2+結合タンパクであるカルモジュリン(CaM)と結合して細胞内Ca2+濃度の上昇を誘起すること、(v)アクチンフィラメントとの結合と細胞膜の損傷(図2④、⑤)などを誘導することが示唆された。上記のIPHではペプチド鎖を3分子導入したが、そのペプチド鎖の数を1、2つに減らしたIPHの設計と合成を行ってそれぞれの抗がん活性を測定した。その結果、カチオン性(塩基性)ペプチドの数と抗がん活性の間に正の相関が観測されることがわかった。

Reason

上記の目的を達成するために、前年度は入手容易なキラルアミンとチオフェンまたはフラン化合物との反応によって、光学活性な多脚アロイルチオウレア配位子を合成し、Ru錯体に導入した。しかし、これらの錯体は有機溶媒中で安定であったものの、水の存在下では分解していることが示唆された。これらの結果はある程度想定されていたので、水溶液中において安定であることが確認されているシクロメタレート型イリジウム(Ir(III))錯体―ペプチドハイブリッド(IPH)に対して、カチオン性ペプチドを1,2,3分子導入した化合物をつくり分け、それぞれのがん細胞に対する抗がん活性を測定、評価することにした。

Strategy for Future Research Activity

前年度における他の研究成果から、IPHによる細胞死は、「Ca2+の小胞体からミトコンドリアへの直接移動に基づくミトコンドリア内Ca2+濃度の上昇によって誘起されるパラプトーシス」であることが強く示唆された。そこで、ウェスタンブロッティング法や、細胞内器官のイメージング試薬との共染色などの実験によって、その詳細なメカニズムを検討する。特に、IPHが細胞内小器官である小胞体からミトコンドリアへの直接的なCa2+輸送を誘起している可能性が考えられるため、それらの細胞内小器官の選択的発光試薬との共染色実験などを行う。また、IPHのターゲット候補分子であるカルモジュリンとの複合体の結晶化とX線結晶構造解析を試みる。
