Outline of Annual Research Achievements

赤痢アメーバの病原機構を理解することを目的として、哺乳動物細胞との相互作用に伴い発動されるAGCK1シグナリングを解明するために、赤血球に対する受容体をシアル酸結合ビーズによる生化学的方法により同定したところ、ガラクトース・Nアセチルガラクトサミン特異的レクチンの重サブユニットの新規タンパク質が同定された。また、貪食に関わるフォスファチジルイノシトール3リン酸(PIP3)の産生に関与するキナーゼを同定した。同時にPIPをオルガネラ間で輸送するPIP輸送タンパク質のゲノムからの同定と基礎的なツール(発現体、発現抑制体)の作成を行った。以上、接着及び咀嚼食に関与するタンパク質のインビトロでの病原性・病害性の関与を発現抑制体・過剰発現体の表現型解析により検証した。

Reason

概ね予定通り進行しているが、COVID-19により若干の計画の遅れを来した。特に長時間の拘束を必要とされるラボでのインビトロライブイメージングに関する計画が若干の遅れをもたらした。

Strategy for Future Research Activity

ラボ内の人員の配置等を工夫することにより、上記の問題点は比較的速やかに解決することが期待される。今後、PIP3の分解に関与するPTENが咀嚼食と貪食に関与することを証明することを目指す。また、PIPをオルガネラ間で輸送するPIP輸送タンパク質の咀嚼食への関与を証明すること目的として、PI3PとPI(3,4)P2の輸送に関与するタンパク質の性状と機能を解析する予定である。
