| Outline of Annual Research Achievements |
人工関節インプラント感染症（prosthetic joint infection: PJI）における自然免疫・オートファジーに関連した生体反応機構の解明を行っている。
昨年は、PJI組織の主要構成細胞のマクロファージ、好中球、線維芽細胞におけるTLR (Toll-like receptor)、NLR (Nod-like receptor)ならびに選択的オートファジー関連分子（WIPI-2）の細胞局在と機能解析を蛍光二重染色によって行った。その結果は、PJI組織のマクロファージ様細胞 (CD68陽性)にWIPI-2陽性像が多くみられた。また、透過型電子顕微鏡による超微細組織・細胞評価でも、同様にマクロファージ様細胞にオートファジーを認めた。
また、比較対象として、変形性関節症、関節リウマチ滑膜組織においてもToll様受容体、Nod様受容体ならびに選択的オートファジー関連分子（WIPI-2）の細胞局在と機能解析を行った。関節リウマチの滑膜組織では、PJI組織と同様に線維芽細胞様細胞 (TEM-1陽性細胞)よりもマクロファージ様細胞 (CD68陽性)にWIPI-2陽性像が多くみられた。変形性関節症の滑膜組織では、関節リウマチ滑膜組織よりオートファージを示す細胞が少なかった。また、疾患活動性や滑膜炎の状態に、オートファジーがどの程度関与するか検討を行っている。
また、他のオートファジーマーカーや自然免疫反応の相互作用をさらに検討していく予定である。

人工関節インプラント感染症（prosthetic joint infection: PJI）組織、関節リウマチ組織、変形性関節症組織において、オートファジーマーカーの検討を行い、有意な結果が得られた。また、電子顕微鏡検査も同時に行い、オートファジーとPJIの関連を調査するための基礎的なデータが得られたため。

| Strategy for Future Research Activity |
人工関節インプラント感染症（prosthetic joint infection: PJI）組織、関節リウマチ組織、変形性関節症組織において、オートファジーマーカーのmRNAをRT-PCRにより解析し、タンパクレベルの解析を免疫ブロット / ELISA法を用いて同定する。また、インプラント動物モデルを確立し、培養細胞系を樹立し、各種培養条件におけるオートファジー関連物質のFACS、RT-PCR、免疫ブロット / ELISA法解析を行う。
