| Outline of Annual Research Achievements |
胸腺退縮は被虐待児の特徴の一つで，虐待ストレスによって胸腺のリンパ球にアポトーシスが誘導されるためと考えられている．そのアポトーシスの過程においてケモカインの関与が示唆されている．そこで，マウスにおけるストレス誘発性胸腺退縮におけるケモカインの役割を解析する．野生型マウスで1日1時間拘束ストレスにさらすと，胸腺が退縮することを明らかにした．CX3CR1ケモカインレセプターに着目し，CX3CR1遺伝子欠損マウスを同様に拘束ストレスに暴露した時の胸腺の変化についてみたところ，CX3CR1遺伝子欠損マウスでは胸腺退縮の程度が軽減していた．また，胸腺細胞のアポトーシスについても，CX3CR1遺伝子欠損マウスではアポトーシス細胞の数が明らか減少していた．副腎皮質ホルモンについては，CX3CR1遺伝子欠損マウスに比べて野生型マウスでは有意に増加していた．すなわち，CX3CR1遺伝子欠損マウスでは副腎皮質ホルモンを抑制することでストレス誘発性胸腺退縮に対して抵抗性を示すことが明らかになった．さら，胸腺におけるケモカイン（CCL2,CCL3, CCL4, CCL5），ケモカインレセプター（CCR2, CCR5）および炎症性サイトカイン（IL-1β，IL-6, IL-12p35，IL-12p40）の遺伝子発現を検討したところ， 野生型マウスとCX3CR1遺伝子欠損マウスの間で差は認められなかった．ストレス応答には視床下部-下垂体-副腎系（hypothalamic-pituitary-adrenal axis；HPA axis）が重要な役割をしていることから，視床下部おけるケモカインレセプターの発現を検討したところ，野生型マウスでは視床下部の神経細胞にCX3CR1発現を認めた．このことから，視床下部の神経細胞におけるCX3CR1発現が，胸腺退縮における重要な役割を果たしていることが示唆された．
