| Outline of Annual Research Achievements |
翻訳アレスト配列をゲノム網羅的に同定するために、コンピュータ解析による配列ビッグデータ横断解析に基づく情報工学的な方法論を確立し、ESUCA法と名付け、論文として発表した(Takahashi et al., BMC Genomics, 2020)。ESUCA法の支援手法として、アレスト配列とタンパク質酵素活性配列のmicro syntenyを活用したクラスタリング手法を開発して、ESUCA法の出力結果をクラスタリングして、アレスト配列候補配列を類似性の高いホモロジーグループ(HG)に分類することが出来た。ESUCA法を5つの植物ゲノム(シロイヌナズナ、イネ、ポプラ、ブドウ、トマト)に応用することで、新規のアレスト配列候補配列を抽出し、100近くの新規HGを発見した。植物に関する11個のアレスト配列候補配列をルシフェラーゼによる実験的に検証することで、少なくとも5個アレスト配列を発見しこの成果を論文として発表することが出来た(Takahashi et al., BMC Genomics, 2020)。4つ動物ゲノム(ヒト、ショウジョウバエ、ゼブラフィッシュ、ニワトリ)に応用することで、1000個近くのアレスト候補配列を抽出した (Takahashi et al., bioRxiv, 2020)。ヒトゲノム上の翻訳アレスト配列候補配列は、web上に公開されたRibo-Seqデータに基づき、可能性が高い候補を選んで実験を行ったところ、7個のアレスト配列候補配列を同定できた。この研究についても、論文を準備中である。

当初は、膨大な候補データが出力され、詳細な検討が困難になったため、遅れが生じたが、様々な工夫をすることで、解決出来た。現在では、予想以上に、良好な結果が得られている。

これまで、進化保存的なAUG型uORFに注目したが、non-AUG型uORFに着目して、より多くの翻訳アレスト配列を見つけることを試みている。
