生体分子機械による革新的動力システムの合理設計

Outline of Annual Research Achievements

本研究では、生体分子機械(微小管・キネシン)における1)集団運動の発現機構、2)集団運動の構造的安定性、3)集団運動のエネルギー収支を微視的および巨視的な観点から統一的に理解することを目的としている。これらの知見をもとに生体分子機械による革新的動力システムの合理的な設計指針を提案する。具体的な研究課題は以下のとおりである。課題①生体分子機械の集団運動を支配する制御因子の探索とその安定性の評価、課題②高速数値シミュレーション技術を用いた生体分子機械の形態予測、課題③生体分子機械による集団運動の熱力学的考察となる。昨年度、課題1)に関しては、実験系で指標とした制御因子(微小管の長さ、曲げ剛性、相互作用の程度)を考慮しながらキネシンを駆動し、開発済みの実時間可視化シミュレーションを用い、数万本規模の微小管の形態予測を行った。集団運動発現に必要となる引力相互作用は微小管間にレナード・ジョーンズ型のポテンシャルを働かせることで導入した。さらにシミュレーションと実験結果を比較しながら制御因子の統一的な理解をはかった。課題2)に関しては、開発済みの力学ストレス印可装置上で集団運動を発現させるとともに、その集団運動に歪、歪速度、伸張パターンを系統的に変化させながら力学的ストレスを印加し、その結果生じる運動特性の変化を相関解析や平均二乗変位量などの解析することで安定性の評価を行った。課題3)に関しては、集団運動する微小管から正味の仕事として取り出せるエネルギー量の見積もりおよび仕事効率を算出することで熱力学的な観点からの考察を行った。正味の仕事として取り出せるエネルギー量は、ATPの加水分解によって得られるエネルギー量をもとに、仕事効率は、秩序パラメーターを全微小管の平均角度を画像解析により求めた。様々な集団運動に関して統計的な解析を行うことで系のエネルギー収支に関しても考察を行った。
