Outline of Annual Research Achievements |
Last year, I did a research project in the interaction of environmental chemicals and thyroid hormones with the nuclear receptor (thyroid hormone receptor and estrogen receptor) or cell surface receptor (integrin and GPER1/GPR30) that regulate cell proliferation and migration. The results showed that thyroid hormones induced cell migration and proliferation through TR-dependent and TR-independent pathways. The thyroid hormone-induced cell migration and proliferation through the activation of integrin that leads to activate the TR-independent pathways. In addition, environmental chemicals such as isoflavones accelerate cell migration through GPER1/GPR30 activation, which leads to activate PI3K/Akt/Rho-GTPase pathways that induced F-actin rearrangement. Gadolinium-based contrast agents (GBCA) also increased cell proliferation and cell migration through integrin activation pathways, which increased the phosphorylation activity of ERK1/2 and Akt. I published two articles in the International Journal of Molecular Science and the Journal of Cellular Physiology. Now, I have one manuscript in submission to Frontiers in endocrinology with the title "Soy isoflavones accelerate glial cell migration via GPER-mediated signal transduction pathway" and one manuscript in preparation as first authors with the title "Gadolinium-based contrast agents induced cell migration via integrin αvβ3 signaling pathway". I also prepared one review article as co-author for an invitation from the International Journal of Environmental Research and Public Health.

1: Research has progressed more than it was originally planned.
Reason
Every fiscal year I always published at least two articles. This year I have two manuscripts in preparation and one in submission. I also have collaboration research with other departments, and it almost finished. Now we discuss how to prepare the manuscript for publication. My thyroid hormones project also practically finished this year. I finished to builts the microscope for live-cell imaging and start to work with this next month. I even begin to make a framework for the thyroid hormones manuscripts and rearrange the data for the figures. Since we faced the coronavirus disease (COVID-19) pandemic for these several months, I almost do all of my work from home. I focused on finishing all of my manuscripts, analyzing my data, and do the programming work like in silico molecular docking study. Even I stay at home, and I still manage to do my work properly. I also discussed it with my supervisor and other colleagues.

Strategy for Future Research Activity |
Next year I need to do several step to finish my project during fellowship. I plan to start to do live-cell imaging for my environmental chemicals and thyroid hormone project. I will conduct the live-cell imaging with co-exposure between environmental chemicals or thyroid hormone with the inhibitors; I will investigate the flow and motion of cells when they migrate or proliferate. I also do the western blotting to examines expression levels of protein and phosphorylation protein induced by environmental chemicals and thyroid hormone exposure. I will continue my work with immunocytochemistry to examines the expression level of the various protein target and discover the downstream pathway mechanism. I also plan to do double staining for flow cytometry to examine the mechanism of thyroid hormone-induced cell proliferation. Then I will analyze the data and start to prepare the manuscript for publication. Hopefully, I can publish four articles for the next year.
