Outline of Final Research Achievements |
Our goal was to investigate molecular subtypes of colorectal precancerous lesions in relation to the microbiome signatures through both translational and basic animal studies. We obtained genomic DNA from clinical samples of colorectal precancerous lesions and developed assays for mutation and methylation analysis. We also examined Fusobacterium nucleatum and Fusobacterium whole species using qPCR but the supported study period terminated before additional experiments.

Academic Significance and Societal Importance of the Research Achievements |
大腸腫瘍初期病変臨床検体の分子異常に関わる細菌叢の同定は、発癌メカニズムの理解、化学予防や新規治療開発の観点より意義はあると考えられる。補助機関終了後も本研究は我々の研究室の研究テーマである「消化器炎症・癌患者の臨床検体を用いたGenetic/epigenetic異常の解析と予後に関する検討」の一端として継続予定であり、科研費により購入した解析試薬を用い収集した大腸腫瘍サンプルに関して、候補遺伝子解析をもとにした、分子サブタイプによる分類、細菌解析として、次世代シークエンサーを用いた網羅的細菌解析を行う予定としている。
