Outline of Annual Research Achievements

二分脊椎症は脊髄内にも傷害がおよび、運動、排尿障害、下肢の変形等が起こる重要な疾患である。しかし、これ まで適切なモデル動物が開発されていなかっ た為に充分な病態解明がなされていない。当研究室では、手術により脊髄を再開裂し、世界で初めて歩行異常を示す二分脊椎モデル動物を効率良く作成すること に成功した。本研究は、神経伝達物質に対する免疫組織化学により上記神経異常に関する論文報告を行いながら、最終的には両神経異常を多光子顕微鏡により可視化、検討し、二分脊椎における歩行障害の原因を解明することが目的である。
以前の研究では抑制ニューロンの変化の指標としてGABA免疫染色を行い、抑制系の減少を確認した。さらにカスパーゼの免疫染色により二分脊椎ヒヨコの脊髄に生直前 から神経障害が起こり始めていることを確認した。同時に運動障害の程度を動画データとして投稿ができるようデジタルカメラで記録した。平成29年度には本モ デルを用い、特に興奮性のグルタミン酸、アセチルコリンと、抑制性のGABA の増減に注目して解析し、孵化後に運動機能が急激に悪化すること、それが孵化後 の抑制神経系障害が原因であることを明らかにし、動物モデル分野では世界的に評価の高いDisease Models & Mechanisms (IF=4.7)に論文をKhanの第一著者論文 として発表した。
一方で、トレーサーを用いた二光子顕微鏡による解析を共同研究者との基礎実験を開始した。さらに、二分脊椎モデルにプロサポシンのペプチド配列の中で神経保護作用を有するサポシンＣの配列から作成した18個の合成ペプチドを実際に二分脊椎に投与した。以前の実験結果から決定した投与量を段階的に変更し、治療効果を検討した。現在のところ明瞭な治療効果が得られており、本年3月の解剖学会で報告し、さらに実験を継続して論文作成を目指している。

Reason

二分脊椎のモデル作成は、ほぼ完璧に出来るようになり、作成効率が安定してきたので、薬効解析等が可能となってきた。二分脊椎モデルにプロサポシンのペプチド配列の中で神経保護作用を有するサポシンＣの配列から作成した18個の合成ペプチドを実際に二分脊椎に投与した。以前の実験結果から決定した投与量を段階的に変更し、治療効果を検討した。現在のところ明瞭な治療効果が得られており、本年3月の解剖学会で報告し、さらに実験を継続して論文作成を目指している。

Strategy for Future Research Activity

サポシンＣの配列から作成した18個の合成ペプチドを実際に二分脊椎に投与したところ、治療効果が期待出来るので、例数を増やし、投与量を変えて実験を進め、論文作成に向けて研究を推進する。出来れば、同時に治療メカニズムに関しても研究し解明したい。

Causes of Carryover

実験に必要な物品費としては1013円は少額であり、使用出来なかった。次年度と合わせてすぐに使用可能な金額である。
