肥満は様々な疾患のリスクになり得ることが知られ、喘息もその一つである。スフィンゴリン脂質の生合成を調節するorosomucoid-like protein 3 (ORMDL3)は小児喘息の発症リスクであることが報告され、また重症喘息のなかに成人発症，非アトピー型、女性、肥満というフェノタイプが存在することも指摘されている。しかしそれらの詳細な分子メカニズムは未だ明らかにされていない。
本研究の目的は、脂肪酸組成の不均衡がどのような機序でアレルギー性気道炎症と関連するのか喘息モデルマウスを用いて検証することである。我々はとくにC12-16の飽和/一価不飽和脂肪酸の伸長活性を有する長鎖脂肪酸伸長酵素 Elongation of long-chain fatty acids family member 6 (Elovl6)の活性とアレルギー性気道炎症に着目し、検討を進めている。
12週齢の野生型C57BL/6系マウス (WT)、Elovl6欠損型C57BL/6系マウス (Elovl6 -/-)に対し、day 0とday 14に100 μgの卵白アルブミン (OVA)を皮下感作し、day 28に10 μgのOVAを経鼻曝露して抗原特異的アレルギー性気道炎症モデルを作製した。昨年までの検討で、OVA免疫Elovl6 -/-群ではWT群と比べて、type2気道炎症とnon-type2気道炎症の両者が増悪することが分かっている。そこで今年度は、ステロイド抵抗性の要因となるnon-type2気道炎症を中心にElovl6の影響について検討を行った。
